Guanidine hydrochloride is a strong chaotropic agent used in biochemistry and molecular biology for denaturation and subsequent refolding of proteins. In addition, this strong denaturant is capable of solubilizing insoluble or denatured proteins, such as inclusion bodies. This process is used as a first step in the restoration of the active form of proteins and enzymes, and urea and dithiothreitol (DTT) may also be necessary for this refolding process. Guanidine hydrochloride is used for RNA isolation to dissociate the nucleoprotein into its nucleic acid and protein components, it is also a ribonuclease inhibitor and highly concentrated solutions (6 – 8 M) are used to denature native globular proteins. Guanidine hydrochloride is believed to disrupt the hydrogen bonds that maintain the unique structure of proteins.
