Seamless Cloning Master Mix is a ready-to-use reaction mixture for seamless cloning reactions. Allows successful assembly of multiple DNA fragments, regardless of fragment length or final compatibility. It has been rapidly adopted by the synthetic biology community due to its ease of use, flexibility and suitability for large DNA constructs. Seamless Cloning Master Mix efficiently joins multiple overlapping DNA fragments in an isothermal single-tube reaction.
The system allows cloning of DNA fragments in virtually any linearized Escherichia. coli, does not require pre-existing recombination sites or additional DNA sequences, and eliminates the need for extensive enzymatic DNA treatments such as restriction and ligation. The enzyme mix provided with Seamless Cloning Master Mix accurately recognizes and assembles DNA fragments that share a final homology of 15-20 base pairs (bp) that can be created by PCR amplification. The end result is a fully sealed double-stranded DNA molecule that can serve as a template for PCR, RCA or a variety of other molecular biology applications, including direct transformation.
